We identified seven <i>drol1<\/i> suppressors across four loci, all encoding subunits of the U5 snRNP. Although AT\u2013AC splicing was partially restored in these suppressors, their phenotypes were almost completely rescued. Artificial introns with either GT\u2013AG or AT\u2013AC termini showed a splicing preference for GT\u2013AG termini in <i>drol1<\/i> and its suppressors. These results suggest that AT\u2013AC introns are spliced by a GT\u2013AG specific spliceosome in the suppressors, with DROL1 influencing U5 snRNP specificity at the 5\u2032 splice site. We propose that mRNAs retaining unspliced AT\u2013AC introns impair translation and produce nuclear signals that contribute to the complex phenotypes seen in <i>drol1<\/i>.<\/p>\n<h4>\u8981\u7d04<\/h4>\n<p>\u307b\u3068\u3093\u3069\u306e\u30a4\u30f3\u30c8\u30ed\u30f3\u306e\u672b\u7aef\u306e\u5869\u57fa\u914d\u5217\u306fGT\u2013AG\u3067\u3042\u308b\u304c\u3001\u4e00\u90e8\u306e\u771f\u6838\u751f\u7269\u306b\u306fAT\u2013AC\u3067\u3042\u308b\u30a4\u30f3\u30c8\u30ed\u30f3\u304c\u5b58\u5728\u3057\u3001\u3053\u308c\u3089AT\u2013AC\u30a4\u30f3\u30c8\u30ed\u30f3\u306e\u30b9\u30d7\u30e9\u30a4\u30b7\u30f3\u30b0\u306f\u30b7\u30ed\u30a4\u30cc\u30ca\u30ba\u30ca\u306e<i>defective repression of OLE3::LUC 1<\/i>\uff08<i>drol1<\/i>\uff09\u5909\u7570\u682a\u4e2d\u3067\u306f\u6291\u5236\u3055\u308c\u3066\u3044\u308b\u3002\u79c1\u305f\u3061\u306f\u3001<i>drol1<\/i>\u5909\u7570\u682a\u306e\u30b5\u30d7\u30ec\u30c3\u30b5\u30fc\u30927\u682a\u5358\u96e2\u3057\u3001\u305d\u308c\u3089\u304c\u3044\u305a\u308c\u3082\u30b9\u30d7\u30e9\u30a4\u30bd\u30bd\u30fc\u30e0\u306eU5 snRNP\u306e\u30b5\u30d6\u30e6\u30cb\u30c3\u30c8\u3092\u30b3\u30fc\u30c9\u3059\u308b4\u3064\u306e\u907a\u4f1d\u5ea7\u306b\u3042\u308b\u3053\u3068\u3092\u793a\u3057\u305f\u3002\u3053\u308c\u3089\u306e\u30b5\u30d7\u30ec\u30c3\u30b5\u30fc\u306f<i>drol1<\/i>\u306e\u8868\u73fe\u578b\u3092\u307b\u307c\u5b8c\u5168\u306b\u6291\u5727\u3057\u3066\u3044\u305f\u304c\u3001AT\u2013AC\u30a4\u30f3\u30c8\u30ed\u30f3\u306e\u30b9\u30d7\u30e9\u30a4\u30b7\u30f3\u30b0\u304c\u90e8\u5206\u7684\u306b\u3057\u304b\u56de\u5fa9\u3057\u3066\u3044\u306a\u304b\u3063\u305f\u3002\u672b\u7aef\u304cGT\u2013AG\u307e\u305f\u306fAT\u2013AC\u306e\u3044\u305a\u308c\u3067\u3082\u30b9\u30d7\u30e9\u30a4\u30b7\u30f3\u30b0\u3055\u308c\u308b\u3088\u3046\u8a2d\u8a08\u3057\u305f\u4eba\u5de5\u30a4\u30f3\u30c8\u30ed\u30f3\u3092\u690d\u7269\u306b\u5c0e\u5165\u3057\u305f\u3068\u3053\u308d\u3001\u305d\u308c\u3089\u306f<i>drol1<\/i>\u304a\u3088\u3073\u305d\u306e\u30b5\u30d7\u30ec\u30c3\u30b5\u30fc\u306b\u304a\u3044\u3066\u307b\u307cGT\u2013AG\u672b\u7aef\u3067\u3057\u304b\u30b9\u30d7\u30e9\u30a4\u30b7\u30f3\u30b0\u3055\u308c\u306a\u304b\u3063\u305f\u3002\u3053\u308c\u3089\u306e\u7d50\u679c\u306f\u3001\u30b5\u30d7\u30ec\u30c3\u30b5\u30fc\u4e2d\u3067\u306fAT\u2013AC\u30a4\u30f3\u30c8\u30ed\u30f3\u304cGT\u2013AG\u7279\u7570\u7684\u30b9\u30d7\u30e9\u30a4\u30bd\u30bd\u30fc\u30e0\u306b\u3088\u3063\u3066\u30b9\u30d7\u30e9\u30a4\u30b7\u30f3\u30b0\u3055\u308c\u3066\u3044\u308b\u3053\u3068\u3092\u793a\u5506\u3057\u3066\u304a\u308a\u3001DROL1\u304cU5 snRNP\u306e5&prime;\u30b9\u30d7\u30e9\u30a4\u30b9\u4f4d\u7f6e\u306e\u7279\u7570\u6027\u306b\u95a2\u308f\u308b\u53ef\u80fd\u6027\u3092\u793a\u3057\u3066\u3044\u308b\u3002AT\u2013AC\u30a4\u30f3\u30c8\u30ed\u30f3\u3092\u4fdd\u6301\u3057\u305f\u672a\u6210\u719f\u306amRNA\u306f\u7ffb\u8a33\u3092\u963b\u5bb3\u3057\u3001\u6838\u5185\u30b7\u30b0\u30ca\u30eb\u3092\u751f\u6210\u3059\u308b\u3053\u3068\u3067<i>drol1<\/i>\u306e\u8907\u96d1\u306a\u8868\u73fe\u578b\u3092\u5f15\u304d\u8d77\u3053\u3057\u3066\u3044\u308b\u3068\u8003\u3048\u3089\u308c\u307e\u3059\u3002<\/p>\n<figure
